Sie spielen eine Rolle bei der Bildung von Zilien und Gei\u00dfeln und bilden die mitotische Spindel w\u00e4hrend der Zellteilung. Als Hauptbestandteil der Zentrosomen sind sie au\u00dferdem an der Organisation der Mikrotubuli beteiligt.<\/p>\n\n\n\n<p>Im Zentrum jeder Zentriole befindet sich das sogenannte &#8222;Cartwheel&#8220;, das aus SAS-6-Proteinen besteht. Diese organisieren sich selbst zu gestapelten Ringpolymeren auf der Oberfl\u00e4che eines Torus, der die Mutterzentriole umgibt, und leiten so Wachstum einer neuen Zentriole ein. Bislang war unklar, wie sich die an sich schraubenf\u00f6rmigen SAS-6-Polymere zu flachen Ringen zusammensetzen und wie die Symmetrie bestimmt wird.<\/p>\n\n\n\n<p>In Zusammenarbeit mit Pierre G\u00f6nczys Gruppe in Lausanne, untersuchten Forscher*innen der MBM-Gruppe die Dynamik des SAS-6 Aufbaus. Mit Hilfe von photothermisch aktivierten Hochgeschwindigkeits-Rasterkraftmikroskopie (photothermally-actuated, off-resonance tapping high-speed atomic force microscopy) zeigten sie, dass die Affinit\u00e4t der SAS-6-Kopfdom\u00e4nen auf einer Oberfl\u00e4che um vier Gr\u00f6\u00dfenordnungen h\u00f6her ist als in L\u00f6sung. Grobe Molekulardynamiksimulationen zeigten au\u00dferdem, dass das Vorhandensein einer Oberfl\u00e4che solche Winkel zwischen den SAS-6-Monomeren beg\u00fcnstigt, die mit der charakteristischen, 9-fachen Symmetrie vereinbar sind. \u00dcberbleibsel der helikalen Eigenschaften des Proteins spiegeln sich in der Ausrichtung der C-terminalen Enddom\u00e4ne wieder, was wahrscheinlich die Ringstapelung unterst\u00fctzt.<\/p>\n\n\n\n<p><a href=\"https:\/\/www.nature.com\/articles\/s41467-021-26329-1\">Die gemeinsamen Ergebnisse wurden k\u00fcrzlich in der Fachzeitschrift &#8222;Nature Communications&#8220; ver\u00f6ffentlicht:<\/a><\/p>\n\n\n\n<p>Niccol\u00f2 Banterle,&nbsp;Adrian P.
